Drug action
Velmanase alfa is an enzyme produced by recombinant DNA technology that provides replacement therapy for the treatment of alpha-mannosidosis, a lysosomal storage disorder caused by deficiency of alpha-mannosidase.
Indications and dose
**Non-neurological manifestations of alpha-mannosidosis (under expert supervision)**
- **Adult** (By Intravenous Infusion): 1 mg/kg once weekly.
**Non-neurological manifestations of alpha-mannosidosis (under expert supervision) for velmanase alfa**
- **Neonate** (By intravenous infusion): 1 mg/kg once weekly.
- **Child** (By intravenous infusion): 1 mg/kg once weekly.
Cautions
Infusion-related reactions Cautions, further information Infusion-related reactions can occur and velmanase alfa should only be administered when appropriately trained staff are available; pre-medication with an antihistamine and/or corticosteroid may prevent subsequent reactions in those who required symptomatic treatment. Patients should be closely monitored for signs of infusion-related reactions during and for at least 1 hour after administration, and managed appropriately-consult product literature. M
Side effects
Common or very common Acute kidney injury; appetite increased; bradycardia; chills; confusion; cyanosis; diarrhoea; dizziness; epistaxis; eye erythema; eye irritation; eyelid oedema; fatigue; feeling hot; fever; gastrointestinal discomfort; headache; hyperhidrosis; hypersensitivity; insomnia; joint disorders; loss of consciousness; malaise; myalgia; nausea; pain; procedural headache; psychotic disorder; reflux gastritis; syncope; tremor; urticaria; vomiting; weight increased Frequency not known Hyperthermia; infusion related reaction
Pregnancy
Avoid unless essential-limited information available. M

Suitable for use in breast-feeding-no information available but absorption by the infant is considered to be minimal. M
