Drug action
Pegcetacoplan is a pegylated peptide molecule, which binds to C3 and C3b complement proteins, thereby inhibiting the complement cascade that leads to haemolysis.
Indications and dose
**Paroxysmal nocturnal haemoglobinuria (initiated by a specialist)**
- **Adult** (By Subcutaneous Infusion): 1080 mg twice weekly, administer on day 1 and day 4 of each treatment week, continue current dose of C5 inhibitor for the first 4 weeks of treatment, for dose adjustments due to lactate dehydrogenase levels-consult product literature.
Contraindications
Patients unvaccinated against Neisseria meningitidis , Streptococcus pneumoniae , and Haemophilus influenzae ; unresolved infection caused by encapsulated bacteria (including Neisseria meningitidis , Streptococcus pneumoniae , and Haemophilus influenzae ) Contra-indications, further information Vaccination against serious infection To reduce the risk of serious infection, it is advised that patients are vaccinated against Neisseria meningitidis , Streptococcus pneumoniae , and Haemophilus influenzae according to local guidelines, at least 2 weeks, but no longer than 2 years, prior to starting treatment, unless the risk of delaying treatment outweighs the risk of developing an infection. If immediate treatment is needed, administer the required vaccines as soon as possible; treat with prophylactic antibiotics until 2 weeks after vaccination. M
Side effects
Common or very common Asthenia; diarrhoea; dizziness; epistaxis; fever; gastrointestinal discomfort; headache; increased risk of infection; myalgia; nausea; pain; sepsis; skin reactions; thrombocytopenia

- Based on in vitro data, pegcetacoplan has low potential for clinical drugdrug interactions.
