Drug action
Ixekizumab is a human monoclonal antibody that binds to interleukin-17A and inhibits the release of pro-inflammatory cytokines and chemokines.
Indications and dose
**Moderate-to-severe plaque psoriasis (under expert supervision),Psoriatic arthritis with concomitant moderate-to-severe plaque psoriasis (under expert supervision)**
- **Adult** (By Subcutaneous Injection): Initially 160 mg for 1 dose, followed by 80 mg after 2 weeks, then 80 mg every 2 weeks for 5 further doses (at weeks 4, 6, 8, 10 and 12), then maintenance 80 mg every 4 weeks, consider discontinuation of treatment if no response after 16-20 weeks.
**Psoriatic arthritis (under expert supervision),Ankylosing spondylitis (under expert supervision),Non-radiographic axial spondyloarthritis (under expert supervision)**
- **Adult** (By Subcutaneous Injection): Initially 160 mg for 1 dose, then maintenance 80 mg every 4 weeks, consider discontinuation of treatment if no response after 16-20 weeks.
**Moderate-to-severe plaque psoriasis (under expert supervision) for ixekizumab**
- **Child 6-17 years (body-weight 25-49 kg)** (By subcutaneous injection): Initially 80 mg for 1 dose, then maintenance 40 mg every 4 weeks, consider discontinuation of treatment if no response after 16-20 weeks.
- **Child 6-17 years (body-weight 50 kg and above)** (By subcutaneous injection): Initially 160 mg for 1 dose, then maintenance 80 mg every 4 weeks, consider discontinuation of treatment if no response after 16-20 weeks.

Contraindications
Active infections (including active tuberculosis); inflammatory bowel disease (discontinue if signs or symptoms develop, or an exacerbation occurs)
Side effects
Common or very common Conjunctivitis; increased risk of infection; nausea; oropharyngeal pain Uncommon Angioedema; inflammatory bowel disease; neutropenia; skin reactions; thrombocytopenia Rare or very rare Anaphylactic reaction Frequency not known Hypersensitivity (occasionally late-onset)
Interactions
**Severe interactions:**

**Other interactions (1):**
- In population pharmacokinetic analyses, clearance of ixekizumab was not affected by concomitant administration of oral corticosteroids, NSAIDs, sulfasalazine, or methotrexate.
