Drug action
Imiglucerase is an enzyme produced by recombinant DNA technology that is administered as enzyme replacement therapy for non-neurological manifestations of type I or type III Gaucher's disease, a familial disorder affecting principally the liver, spleen, bone marrow, and lymph nodes.
Indications and dose
**Non-neurological manifestations of type I Gaucher's disease (specialist use only),Non-neurological manifestations of type III Gaucher's disease (specialist use only)**
- **Adult** (By Intravenous Infusion): Initially 60 units/kg every 2 weeks; maintenance, adjusted according to response, doses as low as 15 units/kg once every 2 weeks may improve haematological parameters and organomegaly.
**Gaucher's disease type I (specialist use only) for imiglucerase**
- **Neonate** (By intravenous infusion): Initially 60 units/kg every 2 weeks, adjusted according to response, doses as low as 30 units/kg once every 2 weeks may be appropriate.
- **Child** (By intravenous infusion): Initially 60 units/kg every 2 weeks, adjusted according to response, doses as low as 30 units/kg once every 2 weeks may be appropriate.
**Gaucher's disease type III (specialist use only) for imiglucerase**
- **Neonate** (By intravenous infusion): Initially 60-120 units/kg every 2 weeks, adjusted according to response.
- **Child** (By intravenous infusion): Initially 60-120 units/kg every 2 weeks, adjusted according to response.
Side effects
Common or very common Angioedema; cough; dyspnoea; hypersensitivity; skin reactions Uncommon Abdominal cramps; arthralgia; back pain; chest discomfort; chills; cyanosis; diarrhoea; dizziness; fatigue; fever; flushing; headache; hypotension; nausea; paraesthesia; tachycardia; vomiting
