Drug action
Givosiran is a small interfering RNA, which reduces production of the enzyme ALAS1 involved in haem synthesis in the liver, thereby reducing accumulation of neurotoxic intermediates that cause acute porphyria attacks and symptoms.
Indications and dose
**Acute hepatic porphyria (initiated under specialist supervision)**
- **Adult** (By Subcutaneous Injection): 2.5 mg/kg once a month.
**Acute hepatic porphyria (initiated under specialist supervision) for givosiran**
- **Child 12-17 years** (By subcutaneous injection): 2.5 mg/kg once a month.
Side effects
Common or very common Fatigue; hypersensitivity; nausea; renal impairment; skin reactions
Pregnancy
Use only if potential benefit outweighs risk-toxicity in animal studies. M
Breast feeding
Avoid-present in milk in animal studies. M
Renal impairment
Monitor renal function during treatment-progression of impairment reported. M
