Atogepant is a calcitonin gene-related peptide (CGRP) receptor antagonist which inhibits the function of CGRP, thereby preventing migraine attacks.
Drug action
Atogepant is a calcitonin gene-related peptide (CGRP) receptor antagonist which inhibits the function of CGRP, thereby preventing migraine attacks.
Indications and dose
**Prophylaxis of episodic or chronic migraine [in patients who have at least 4 migraine days per month]**
- **Adult** (By Mouth): 60 mg once daily.
Side effects
Common or very common Appetite decreased; constipation; drowsiness; fatigue; nausea; weight decreased
Interactions
**Severe interactions:**
- CYP3A4 Inhibitors Co-administration of AQUIPTA with itraconazole, a strong CYP3A4 inhibitor, resulted in a significant increase in exposure of atogepant in healthy subjects.
- OATP Inhibitors Co-administration of AQUIPTA with single dose rifampicin, an OATP inhibitor, resulted in a significant increase in exposure of atogepant in healthy subjects.
**Other interactions (2):**
- The recommended dosage of AQUIPTA with concomitant use of strong CYP3A4 inhibitors (e.g., ketoconazole, itraconazole, voriconazole, clarithromycin, telithromycin, indinavir, nelfinavir, ritonavir,...
- The recommended dosage of AQUIPTA with concomitant use of strong OATP inhibitors (e.g., rifampicin, atazanavir, ritonavir, tipranavir, ciclosporin, telmisartan) is 10 mg once daily.
Pregnancy
Avoid (toxicity in animal studies). M
Breast feeding
Avoid (present in milk in animal studies). M
Hepatic impairment
Avoid in severe impairment (risk of increased exposure). M
Renal impairment
Dose adjustments Reduce dose to 10 mg once daily if creatinine clearance less than 30 mL/minute. M See Prescribing in renal impairment .
